Poor engine idling/misfiring
Problem:
If you are having any problems with the engine misfiring and poor engine idling, this will be caused by a problem with either the throttle body, or MAF (Mass Air Flow) sensor. These can become clogged up with carbon, resulting in the above problems. This can also result in a lack of power, when you try to accelerate. Is the problem in the video below anything like yours?
Solution:
You can remove the MAF sensor and clean it with some MAF sensor cleaner. Your best option is to clean the throttle body at the same time, with some throttle body cleaner and a tooth brush. Once they are both cleaned out, your problem should be resolved.

Rear brake calliper problem
Problem:
Can you hear a clicking noise, as you pull away or stop the car? This will be most noticeable when moving slowly and when you first apply the brakes. This happens due to a known issue with the brake callipers moving around and hitting their stops. There is no direct fix for this problem, as it is a design fault, which happens on several other Toyota/Lexus vehicles.
Solution:
As mentioned above, there is no direct fix for this issue and it is nothing to worry about. You could try packing something into the gap, in order to stop them moving around.

Heavy clutch wear
Problem:
The IS300 is known for being heavy on the clutch. If you have found that the biting point on the clutch is very high, then this is an indication that the clutch is about to fail.
Solution:
